چکیده

Isothiocyanates (ITCs), the biologically important glucosinolate breakdown products, can present health-promoting effects, play an important role in plant defense and affect plant cellular mechanisms. Here, we evaluated the biological effects of ITCs on Arabidopsis thaliana by assessing growth parameters after long-term exposure to low concentrations of aliphatic and aromatic ITCs, ranging from 1 to 1000 µM. Treatment with the aliphatic allylisothiocyanate (allyl-ITC) led to a significant reduction of root length and fresh weight in a dose-dependent manner and affected the formation of lateral roots. To assess the importance of a hormonal crosstalk in the allyl-ITC-mediated growth reduction, the response of auxin and ethylene mutants was investigated, but our results did not allow us to confirm a role for these hormones. Aromatic ITCs generally led to a more severe growth inhibition than the aliphatic allyl-ITC. Interestingly, we observed a correlation between the length of their side chain and the effect these aromatic ITCs caused on Arabidopsis thaliana, with the greatest inhibitory effect seen for 2-phenylethyl-ITC. Root growth recovered when seedlings were removed from exposure to ITCs.

Glucosinolates, isothiocyanates and indoles

Although isothiocyanate and indole compounds derived from cruciferous vegetables appear to be structurally unrelated, they are both derived from glucosinolates. These sulfur-containing glycosides are found in the order Capparales, which includes the large family Brassicaceae. The glucosinolate molecule comprises a common glucone moiety and a variable aglucone side-chain, derived from one of sev...

Identification of Arabidopsis mutants with altered glucosinolate profiles based on isothiocyanate bioactivity

Glucosinolates are a diverse class of nitrogenand sulfur-containing secondary metabolites. They are rapidly hydrolyzed on tissue disruption to a number of biologically active compounds that are increasingly attracting interest as anticarcinogenic phytochemicals and crop protectants. Several glucosinolate-derived isothiocyanates are potent chemopreventive agents that favorably modulate carcinoge...

Functional detection of chemopreventive glucosinolates in Arabidopsis thaliana.

Natural isothiocyanates, derived from glucosinolates by myrosinase-catalyzed hydrolysis, are potent chemopreventive agents that favorably modify carcinogen metabolism in mammals by inhibiting metabolic activation of carcinogens and/or by inducing carcinogen-detoxifying enzymes. Methylsulfinylalkyl isothiocyanates are potent selective inducers of mammalian Phase 2 detoxification enzymes such as ...

Study of the role of antimicrobial glucosinolate-derived isothiocyanates in resistance of Arabidopsis to microbial pathogens.

Crude aqueous extracts from Arabidopsis leaves were subjected to chromatographic separations, after which the different fractions were monitored for antimicrobial activity using the fungus Neurospora crassa as a test organism. Two major fractions were obtained that appeared to have the same abundance in leaves from untreated plants versus leaves from plants challenge inoculated with the fungus ...

A plastidic transporter involved in aliphatic glucosinolate biosynthesis.

The distinctive flavors and anticarcinogenic properties of cruciferous vegetables are largely due to the presence of glucosinolates. These sulfur-rich metabolites are derived from one of several amino acids via three stages: (1) elongation of the amino acid side chain; (2) formation of the core glucosinolate skeleton; and (3) secondary modification of the side chain (reviewed in Grubb and Abel,...
